Application
The COVID-19 Antigen Test Kit series is based on a solid-phase immunochromatographic assay designed for the rapid and qualitative detection of antigens from the 2019 Novel Coronavirus in human nasal, saliva, or nasopharyngeal samples. This test provides only a preliminary result. Therefore, any reactive specimen detected by the COVID-19 Test Kit (Colloidal Gold Method) must be confirmed using alternative testing methods and evaluated alongside clinical findings.
Principle
The COVID-19 Test Kit (Colloidal Gold Method) is a colloidal gold immunochromatographic assay that detects the nucleocapsid protein on the surface of the COVID-19 virus.
The test utilizes a nitrocellulose membrane strip with immobilized COVID-19 (SARS-CoV-2) antibodies at the test line (T) and goat anti-mouse IgG antibodies at the control line (C). The conjugate pad contains colloidal gold particles bound to another COVID-19 (SARS-CoV-2) antibody and mouse IgG-gold conjugates.
When the sample mixed with buffer is added to the sample well, any COVID-19 antigen present binds to the COVID-19 antibody conjugate, forming an antigen-antibody complex. This complex migrates along the membrane by capillary action. Upon reaching the test line (T), the complex binds to the immobilized antibodies, forming a burgundy-colored band that indicates a reactive (positive) result. The absence of a burgundy band at the test line indicates a non-reactive (negative) result.
The test also includes an internal control line (C) that should always display a burgundy-colored band formed by the immunocomplex of goat anti-mouse IgG and mouse IgG-gold conjugate, confirming that the test is working properly.
If no control line appears, the test result is invalid, and the specimen must be retested with a new device.
