This reagent is used for the qualitative detection of acrosomal enzyme SP10 protein in human semen by immunochromatography. Semen samples are first diluted and added to the test device. If SP10 protein is present, it binds with colloidal gold-labeled antibodies to form a complex that moves along the strip and produces a red band at the test line (T), indicating a positive result. If no SP10 protein is detected or the concentration is below the reference threshold, no red band appears at the test line, indicating a negative result. A red band at the control line (C) must always appear, confirming sufficient specimen volume and proper test performance. Absence of the control line renders the result invalid.

According to the [WHO Manual of Laboratory Testing of Human Semen] (5th edition), the main factors affecting male infertility are: semen volume, total sperm, sperm concentration, liquefaction time, semen PH value, sperm normal form rate, etc., among which sperm concentration detection is an important index to measure male infertility.

Test Procedure
Allow test cassette, Antigen extraction buffer to equilibrate to room temperature prior to testing.
1. Open the aluminum foil bag and take out the test card.
2. Put the test card on a horizontal table, unscrew the top cap of the diluted sample, and slowly add 3 drops (about 100μL) of the pre-treated semen sample to the center of the sample hole;
3. Start timing, the reaction results should be observed within 10-15minutes, and the results are invalid after 15minutes.
